Abstract:
A sensitive method based on capillary electrophoresis(CE) with end-column amperometric detection( AD) has been developed for the chiral separation and determination of sotalol enantiomers. Factors affecting the chiral CE separation of the analytes, such as working potential, acidity, concentration of running buffer and the chiral selector, injection and applied voltage, were extensively investigated. The working electrode operated in the wall-jet configuration was a carbon disk electrode(vs. Ag/AgCl). Under the optimum conditions, baseline separation of the enantiomers could be accomplished, and excellent linearity was observed between peak current and concentration of enantiomers in the range from 5 mu g/L to 500 mu g/L. The limits of detection(S/N = 3) were 2.0 and 1.9 mu g/L, respectively. The proposed method has been applied to the analysis of sotalol in human serum samples with satisfactory results.
