Abstract:

The bacterial species composition and distribution in the intestines of large yellow croaker (Pseudosciaena crocea) during iced storage were investigated by both culture-dependent method and culture-independent method of polymerase chain reaction and denaturing gradient gel electrophoresis(PCR-DGGE). Eleven different bacterial species were identified from spoiage fish samples, including Shewanella, Pseudomonas, Aeromonas, Providencia, Morganella, Bro-chothrix, Acinetobacter, Enterococcus faecium, Escherichia coli, Firmicute and Psychrobacter. Shewanella and Pseudomonas were dominant bacterial groups. The principal component analysis (PCA) of DGGE fingerprint showed that bacterial flora of flesh fish intestines was similar with spoilage one, and Pseudomonas was highly associated with the flora. The proportion of Shewanella increased in bacterial flora after storage obviously. The dominant bacterial speices could been identified by both culture-dependent and PCR-DGGE methods. PCR-DGGE was used to investigate the bacterial diversity and population shifts among samples from different storage times more effectively.
