Abstract:

利用基因组重排技术对产纤维素酶菌株里氏木霉T306进行育种研究,考察原生质体制备、再生及融合条件的影响因素.研究表明,里氏木霉原生质体制备和再生的最佳条件是:3.5%溶壁酶,菌丝菌龄22 h,酶解时间4h,酶解温度30℃,采用pH 5.0磷酸氢二钠-柠檬酸缓冲液,以高渗筛选培养基为再生培养基,再生培养基中渗透压稳定剂为0.8 mol/L NaCl.在此条件下,里氏木霉原生质体量达1.44×107个/mL,再生率为2.64%.融合试验结果表明,将紫外照射90 s和微波辐射200 s两种方式灭活的原生质体融合,在30% PEG、30 mmol/LCa2+、30℃条件下处理12 min,原生质体融合率达0.5%.经两轮基因组重排试验,获得一株遗传稳定,产酶能力较强的融合子G2-0832,其内切葡聚糖酶活力达86.40 U/mL,较出发菌株提高了57.08%.
