Imidacloprid-induced lung injury in mice: Activation of the PI3K/AKT/ NF- κB signaling pathway via TLR4 receptor engagement SCIE

Abstract :

Significant impairment of pulmonary function has been demonstrated through long-term exposure to neonicotinoid insecticides, such as imidacloprid (IMI). However, the underlying mechanisms of lung injury induced by IMI remain unclear. In this study, a mouse model of IMI-induced pulmonary injury was established, and the toxicity and lung damage were assessed through mouse body weight, organ index, hematological parameters, and histopathological analysis of lung tissues. Furthermore, metabolomics and transcriptomics techniques were employed to explore the mechanistic aspects. Results from the toxicity assessments indicated that mouse body weight was significantly reduced by IMI, organ index was disturbed, and hematological parameters were disrupted, resulting in pulmonary injury. The mechanistic experimental results indicate that the differences in metabolites and gene expression in mouse lungs could be altered by IMI. Validation of the results through combined analysis of metabolomics and transcriptomics revealed that the mechanism by which IMI induces lung injury in mice might be associated with the activation of the TLR4 receptor, thereby activating the PI3K/AKT/ NF- kappa B signaling pathway to induce inflammation in mouse lungs. This study provided valuable insights into the mechanisms underlying IMI-induced pulmonary damage, potentially contributing to the development of safer pest control strategies. The knowledge gained served as a robust scientific foundation for the prevention and treatment of IMI-related pulmonary injuries.

Abstract :

Sample preparation is crucial in analytical chemistry, impacting result accuracy, sensitivity, and reliability. Solid-phase separation media, especially adsorbents, are vital for preparing of liquid and gas samples, commonly analyzed by most analytical instruments. With the advancements in materials science, covalent organic frameworks (COFs) constructed through strong covalent bonds, have been increasingly employed in sample preparation in recent years. COFs have outstanding selectivity and/or excellent adsorption capacity for a single target or can selectively adsorb multiple targets from complex matrix, due to their large specific surface area, adjustable pore size, easy modification, and stable chemical properties. In this review, we summarize the classification of COFs, such as pristine COFs, COF composite particles, and COFs-based substrates. We aim to provide a comprehensive understanding of the different classifications of COFs in sample preparation within the last three years. The challenges and development trends of COFs in sample preparation are also presented.

Abstract :

Continuously promoting new curriculum standards is a key aim of the Ministry of Education of China. With this in mind, this paper introduces a comprehensive teaching experiment for undergraduate instrumental analysis courses that aims to improve students' material-preparation and instrumental-analysis skills through practice. Herein, a covalent organic framework-based core-shell stationary phase material (SiO2@COFTTA-DHTA) is prepared via a one-pot method and characterized in detail, after which its chromatographic properties are evaluated by high performance liquid chromatography (HPLC). The experimental process includes material synthesis and characterization, as well as studying the chromatographic-retention behavior and chromatographic-separation performance of the material. By the combining theoretical science and experimental teaching, this experiment not only deepens students' understanding of the properties of functional materials and their applications, but also improves their experimental-design and critical-thinking skills. This experiment not only cultivates students' interests in scientific research, but also exercises their experimental, operational, innovative-thinking, and practical abilities, while concurrently enhancing their sense of social responsibility and historical mission, thereby delivering the all-round educational goals of experimental teaching.

Abstract :

Triclosan (TCS), triclocarban (TCC), and chlorophenols (CPs) are broad-spectrum antibacterials widely used in dermatological and oral hygiene products, which could induce severe liver and intestine injuries. Hence, it is essential to establish a rapid and sensitive method to monitor TCS, TCC, and CPs in various organisms. In this work, fluorine-functionalized covalent organic framework (COF-F) was prepared by using 4,4 ',4"-(1,3,5triazine-2,4,6-triyl)tri-aniline and 2,3,5,6-tetrafluoroterephthalaldehyde as two building units and employed as a solid phase microextraction (SPME) probe for the extraction of TCS, TCC and CPs. The COF-F possessed excellent hydrophobicity, a large specific surface area (1354.3 m2 g-1) and high uniform porosity (3.2 nm), which facilitated high selectivity and adsorption properties towards TCS, TCC, and CPs. Therefore, the as-prepared COF-F-SPME in combination with electrospray ionization mass spectrometry has been developed to provide fast and ultrasensitive detection of TCS, TCC, and CPs in biological samples. The established method demonstrated satisfactory linear ranges (0.01-100.00 mu g L-1) and low limits of detection (0.003-0.040 mu g L-1) for TCS, TCC and CPs. The developed method could be successfully applied to detect TCS, TCC and CPs in the liver and kidney tissues of mice, demonstrating the potential for the detection of chlorinated aromatic pollutants in the biological samples.

Abstract :

Solid-phase microextraction (SPME) coupled with electrospray ionization mass spectrometry (ESI-MS) was developed for rapid and sensitive determination of endogenous androgens. The SPME probe is coated with covalent organic frameworks (COFs) synthesized by reacting 1,3,5-tri(4-aminophenyl)benzene (TPB) with 2,5-dioctyloxybenzaldehyde (C8PDA). This COFs-SPME probe offers several advantages, including enhanced extraction efficiency and stability. The analytical method exhibited wide linearity (0.1-100.0 mu g L-1), low limits of detection (0.03-0.07 mu g L-1), high enrichment factors (37-154), and satisfactory relative standard deviations (RSDs) for both within one probe (4.0-14.8%) and between different probes (3.4-12.7%). These remarkable performance characteristics highlight the reliability and precision of the COFs-SPME-ESI-MS method. The developed method was successfully applied to detect five kinds of endogenous androgens in female serum samples, indicating that the developed analytical method has great potential for application in preliminary clinical diagnosis.

Abstract :

Frequent oil spills cause substantial economic losses and pose a significant threat to human life. Urgent measures must be taken to develop a highly effective approach to mitigating oil spills and minimizing their potential consequences. Herein, we synthesized MS@COFs-F, fluorinated covalent organic framework-decorated superhydrophobic sponges, to clean up crude oil spills efficiently. The synthesis method involved a simple and effective room-temperature approach, utilizing 2,4,6-tris(4-aminophenyl)-1,3,5-triazine (TAPT) as the amine monomer and binding it with three aldehyde monomers of varying fluoride content. The MS@COFs-F with the highest fluorine content, known as MS@COFs-F4, exhibited low surface energy, superhydrophobicity (water contact angle of 155.7 degrees), a large specific surface area of 180 m(2)/g, and exceptional chemical, mechanical, and thermal stability. MS@COFs-F4 demonstrated remarkable effectiveness in adsorbing crude oils and organic solvents of diverse viscosities, exhibiting an adsorption capacity between 68.5 and 125.9 g/g, achieving an oil-water separation efficiency exceeding 97.0 %. Additionally, MS@COFs-F4 showcased self-cleaning and flame-retardant properties, further enhancing its suitability for real-world applications. MS@COFs-F4 holds the potential to offer a novel, safe, and effective approach in addressing oil spill incidents.

本实验旨在通过分散固相萃取-液相色谱-三重四级杆质谱法分析环境水样中的卤代醌类化合物.实验选取了5种卤代醌类化合物作为目标物,采用阳离子共价有机骨架材料作为固相吸附剂,从水样中富集目标物.随后,使用液相色谱-三重四级杆质谱法对富集的化合物进行定性和定量分析.实验考察了吸附剂用量、富集时间、洗脱时间、洗脱溶剂种类、洗脱溶剂酸度和洗脱体积等条件对实验结果的影响,并建立了一种基于分散固相萃取的新方法.该方法在0.1-100 ng∙L-1范围内显示出较好的线性关系(相关系数R2≥0.9924),检出限可达到0.03-0.65 ng∙L-1,并成功应用于分析自来水样品中卤代苯醌类化合物.通过这个实验,学生可以了解基本的样品前处理技术,并提高实验技能、研究和创新思维,增强解决问题的能力.

本实验旨在设计新型固定相以实现各种分析物的高效分离并应用于教学实验.在二氧化硅微球的表面修饰上共价有机框架,合成具有核壳结构的复合材料(SiO2@COFETTA-Td),并将其作为混合模式固定相用于高效液相色谱分离.得益于共价有机框架的优异性质,SiO2@COFETTA-Td填充柱在分离烷基苯类、苯胺类和磺胺类等物质中表现出高柱效和优异的分离度,并成功应用于邻苯二甲酸酯类、抗癫痫药物和PM2.5中多环芳烃类物质的分离,这进一步证明SiO2@COFETTA-Td固定相在复杂生物样品的分离分析潜力.通过这个实验,学生可以熟练掌握高效液相色谱的操作,加深巩固色谱相关理论,了解核壳型固定相分离原理并提高他们的科研思维和创新能力.

Here, an imine-linked-based spherical covalent organic framework (COF) was prepared at room temperature. The as-synthesized spherical COF served as an adsorbent in dispersive solid-phase extraction (dSPE), by its virtue of great surface area (1542.68 m(2)/g), regular distribution of pore size (2.95 nm), and excellent stability. Therefore, a simple and high-efficiency dispersive solid phase extraction method based on a spherical COF coupled with high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) was established to determine aryl organophosphate esters in biological samples. This approach displayed favorable linearity in the range of 10.0-1000.0 ng/L (r > 0.9989), a high signal enhancement factor (58.8-181.8 folds) with low limits of detection (0.3-3.3 ng/L). Moreover, it could effectively eliminate complex matrix interference to accurately extract seven aryl organophosphate esters from mouse serum and tissue samples with spiked recoveries of 82.0%-117.4%. The as-synthesized spherical COF has been successfully applied in sample preparation. The dSPE-HPLC-MS/MS method based on a spherical COF has potential application to study the pollutants' metabolism in vivo.
