Abstract :
Developing a functional media for precise identification of trace shellfish toxin would underpin the effective assessment of marine pollution. Herein, a novel monolithic column with a dual-mode strategy integrating antifouling and aptamer bionic affinity recognition was proposed for online specific identification of the marine toxin okadaic acid (OA). The zwitterionic monomer 2-(methacryloyloxy)ethylphosphorylcholine (MPC) and aptamers were synergistically employed to enable efficient reduction of matrix interferences and selective capture of target OA. Preparation optimization, characterization, and fouling-resistant mechanism of the dualmode bionic monolith were evaluated. The zwitterion phosphorylcholine MPC introduced into the monolith significantly improved the fouling resistance to biomass substrates, meanwhile the aptamers were able to provide a high specific recognition capacity. Coupled with LC-MS, the as-prepared monolith provided an effective approach for highly selective and sensitive identification of OA. Good recovery yields of over 90 % in shellfish tissue extracts and human serum were achieved with a sensitive limit of detection (LOD) as low as 0.1 ng/mL, as well as excellent specificity and low interference from proteins, fatty acids and analogues. Applied to popular shellfishes (such as clams, mussels, and oysters) and serum samples, trace OA toxin was accurately distinguished and quantified with satisfactory recoveries as 93.8 +/- 2.2 % - 99.9 +/- 1.9 % (n = 3). Compared to the traditional HLB cartridge and other materials in the LC-MS method, the resulting anti-fouling aptamer monolith provided a more advanced online analysis mode with higher sensitivity and better resolution of OA in biological samples. It might provide an attractive access to an online bionic recognition platform with LC-MS for efficient, antiinterference and sensitive specific detection of trace marine toxin OA in biological samples.

Abstract :
Surface engineering of covalent organic frameworks (COFs) represents a highly promising strategy for achieving superior capture capacity towards environmental trace pollutants. Herein, the three-dimensional covalent organic frameworks (3D-COFs) with tunable crystallinity were synthesized and optimized via a simple solvent ratio adjustment, demonstrating an exceptional capture capacity of polychlorinated biphenyls (PCBs). By adjusting the ratio of solvents (1,4-dioxane and toluene), a functional 3D COF (dia-c5 TPA-TAM-COF) was discovered, providing an ideal approach to transform an amorphous compound into a functional platform with a significant increase in crystallinity, surface area and pore volume for PCBs capture. The adsorption features and analytical performance of PCBs were evaluated, and a good performance was achieved with high enrichment factors as 6973 ∼ 11,015. Optimization of solid phase microextraction (SPME) using the dia-c5 TPA-TAM-COF as the coating was investigated, and the method was verified for sensitive quantification of PCBs by using GC–MS, achieving the sensitive limits of detection (LODs) of 0.028 ∼ 0.0263 ng/L. Good reproducibility and robustness of the analytical performance of PCBs using dis-c5 was achieved, with the RSDs of intra-day (n = 3), inter-day (n = 6) and fiber-to-fiber (n = 3) of 0.7 ∼ 4.7 %, 1.8 ∼ 4.6 % and 2.0 ∼ 3.9 % respectively. The recoveries of PCBs in the soil fortified samples were obtained as 85.3 ∼ 101.9 % and 86.3 ∼ 108.8 %, respectively, as well as high thermal stability and long lifetime (keep on going more than 150 cycles). This study demonstrates that a simple protocol of adjusting the solvent content was capable of tailoring high crystallinity and suitable interpenetration degree of TPA-TAM-COFs for high performance target capture of PCBs in environmental soils. © 2025

Abstract :
Accurate quantitation of trace-level ATP biomarkers often suffers from interference of sample matrix and remains a challenge. Herein, a novel magnetic biological nanosensor (MBNS) integrating tetrahedral framework nucleic acid (tFNA)-confined aptamers and ultrasensitive laser-induced fluorescence (LIF) was tailored and evaluated. The fluorescent aptamer nanoprobe hybridized with FAM-labelled cDNA was particularly incorporated into the inner cavity of tFNA, enabling a dual-mode mechanism that integrates molecular size selection and aptamer affinity recognition. By this strategy, the benefits of tFNA molecular sieve, the super sensitive properties of LIF, and the specific recognition of "aptamer gene codes" were integrated, as triggered the superior response of ATP. The structures, properties and reaction mechanism of tFNA, as well as tFNA modified with aptamer probe at different sites, were characterized in detail. A high level of anti-interference was achieved, shielding against nonspecific adsorption even in matrices with high protein and analogue content, 10(3)similar to 10(5) times higher than ATP. The release of fluorescent beacon from aptamer probe was facilely driven by specific binding of ATP, and an ultra-sensitive detection of 75 pmol/L ATP was obtained without amplification. It was 2-3 orders of magnitude more sensitive than most fluorescent aptasensors. Applied to serum samples, the stability and reproducibility of the resultant aptasensor have been validated with the intra- and inter-assay RSD in 1.6-6.4 % and -1.5-8.4 %, respectively. Acceptable recoveries of 96.5-106.8 % were achieved, consistent with the LC-MS method. It was expected to open a simple and efficient protocol for highly specific and ultrasensitive ATP detection in clinical diagnostics.

Okadaic acid (OA) biotoxin acts a well-established inhibitor of protein phosphatase even a tumor promoter of human being, arouse great attention in safety monitoring. However, the powerful and convenient nanosensing technologies for addressing the demands such as rapidity, high sensitivity, and stability in the in-field test of OA shellfish toxin is still scarce. Herein, a high-performance magnetic biometric nanosensor (MBNS) integrating oriented aptamers and ultrasensitive laser-induced fluorescence (LIF) was firstly proposed for the in-field detection of trace OA in seafoods. High-density aptamers hybridized with FAM-labeled cDNA were tethered to the surface of AuNPs on magnetic MIL-101@Fe3O4, and then finely regulated by mercaptohexyl alcohol (MCH) to be orderly assembled, as was successfully utilized to engineer an active biological nanosensor for highly specific recognition of OA. Aptamers anchored on magnetic Fe3O4@MOF@AuNPs activate a biometric micro -reactor of OA, in which the superior LIF properties, conformation regulation of aptamer, and the specific recognition using aptamer genes were adopted. The magnetic nanosensor with an excellent specificity and super sensitivity for OA analysis was achieved within 20 min. Moreover, the content of captured OA could facilely be recorded by measuring the fluorescence intensity, and the limit of detection (LOD) and limit of quantitation of OA (LOQ) reached 0.015 and 0.050 ng/mL respectively, which was far better than most aptamer-based biometric sensing methods. The feasibility for accurate test of trace OA toxin in the fortified shellfish samples was validated with the recovery yields of 88.2-107.5% and RSD of 0.5-7.6%, respectively. The result demonstrated that the oriented-aptamer encoded MNS had significant practical values in rapid and ultrasensitive detection of OA biotoxin and the related safety applications.

Abstract :
Androgenetic alopecia (AGA) is a chronic and progressive form of hair loss characterized by vascular degeneration in the perifollicular microenvironment, leading to cell apoptosis and eventual loss of hair follicles (HFs). Traditional therapeutic formulations, such as Minoxidil (MXD) tincture, have limitations in reshaping the perifollicular microenvironment and exhibit limited effectiveness. Here, we report a multi-synergistic therapeutic platform for high-performance hair regeneration therapy. The platform combines microneedle (MN) patches loaded with MXD-encapsulated nanostructured lipid carriers (MXD-NLC-MNs) and cold atmospheric plasma (CAP). The MNs' mechanical strength enables efficient transdermal delivery of MXD to the targeted dermal papilla cells, promoting cell proliferation. Furthermore, in collaboration with MXD, the mechanical stimulation exerted by MN application synergistically upregulates the expression of vascular endothelial growth factor, leading to neoangiogenesis. Meanwhile, the transient microchannels in the skin created by MNs facilitate the transdermal delivery of CAP-generated nitric oxide (NO) to the sites of HF lesions, whereby the synergistic interaction between MXD and NO boosts perifollicular vasodilation. Consequently, the perifollicular microenvironment can be effectively reshaped to accelerate hair regeneration in AGA murine models. This multi-synergistic combination therapy strategy would hold great promise for effectively treating AGA and promoting hair regrowth.

The differentiation of 3-AminoPyrrolidine (3AP) and its derivative isomers is significant due to their use as intermediates in drug synthesis. However, the separation of these isomers, which exhibit both chiral and positional isomerism, has consistently posed challenges. In this study, an efficient analytical method was developed to discrimination between the four isomers of R/S-I/N-Boc-3-AminoPyrrolidine (R/S-l/N-Boc-3AP) and four groups of derived chiral isomers, namely R/S-N-Methyl-3-AminoPyrrolidine (R/S-N-Me-3AP), R/S-N-Ethyl-3AminoPyrrolidine (R/S-N-Et-3AP), R/S-1-Bzl-3-AminoPyrrolidine (R/S-1-Bzl-3AP), and R/S-1-Cbz-3AminoPyrrolidine (R/S-1-Cbz-3AP). The discrimination was achieved by utilizing natamycin (Nat) as a differentiation agent and 2/3/4-aminobenzenesulphonic acid (2/3/4-ABSA) as ligand. The facile discrimination of the four positional isomers of R/S-1-Boc-3AP and R/S-N-Boc-3AP was achieved using ion mobility spectrometry (IMS) based on their formed diastereomeric complexes [Nat + (Boc-3AP)2 + (2-ABSA)2 + H]+. The isomers were also studied via tandem mass spectrometry (MS/MS) using the formed ternary complexes, different fragment ions were observed for l-Boc-3AP and N-Boc-3AP, but the R/S enantiomers cannot be discriminated either. In addition, the enantiomers R/S-l-Bzl-3AP and R/S-l-Cbz-3AP were differentiated based on mobility using [Nat + (R/Sl-Bzl-3AP) + (3-ABSA)2 + H]+ and [Nat + (R/S-l-Cbz-3AP) + 2-ABSA + H]+; however for the small enantiomeric molecules of R/S-N-Me-3AP and R/S-N-Et-3AP, chiral discrimination was not achieved via the ternary complexes of Nat + R/S-N-Me-3AP-ABSA and Nat + R/S-N-Et-3AP-ABSA. Importantly, the separation was greatly improved by the coordinated alkali metal ions, where the separation resolution (Rp-p) increased with the radius of the metal ions. Finally, quantitative analysis and method validation were carried out for the four Boc-3AP isomers using both MS/MS and IMS-MS. Overall, the proposed method opens possibilities for the rapid discrimination and quantification of isomers exhibiting both chiral and positional isomerism in MS/MS and IMS-MS analyses.

The advancement in miniaturized Raman spectrometers, coupled with the single-molecule-level sensitivity and unique fingerprint identification capability of surface-enhanced Raman scattering (SERS), offers great potential for point-of-care testing (POCT). Despite this, accurately quantifying analyte molecules, particularly in complex samples with limited sample volumes, remains difficult. Herein, we present a versatile and reusable SERS microplatform for highly sensitive and reliable quantitative detection of adenosine triphosphate (ATP) in biological fluids. The platform utilizes gold-Prussian blue core-shell nanoparticles modified with polyethyleneimine (Au@PB@PEI NPs), embedded within gold nanoparticle-immobilized capillary-based silica monolithic materials. PB acts as an internal standard, while PEI enhances molecular capture. The periodic, bimodal porous structure of the silica monolithic materials provides uniform and abundant sites for nanoparticle attachment, facilitating rapid liquid permeation, intense SERS enhancement, and efficient enrichment. The platform regulates ATP capture and release through magnesium ions in the liquid phase, eliminating matrix interferences and enabling platform reuse. Integrating efficient molecular enrichment, separation, an interference-free internal standard, a liquid flow channel, and a detection chamber, our platform offers simplicity in operation, exceptional sensitivity and accuracy, and rapid analysis (similar to 10 min). Employing PB as an internal calibration standard, ratiometric Raman signals (I-732/I-2123) facilitate precise ATP quantification, achieving a remarkable limit of detection down to 0.62 pM. Furthermore, this platform has been proven to be highly reproducible and validated for ATP quantification in both mouse cerebrospinal fluid and human serum, underscoring its immense potential for POCT applications.

Sensitive detection of dopamine (DA) is essential for the diagnosis of nervous system diseases. Herein, a nanozyme-based colorimetric platform for DA determination was proposed. Using an imine-type covalent organic framework (COF-TD) as a host matrix, a nanozyme (AgNPs@COF-TD) was fabricated by anchoring silver nanoparticles (AgNPs) on the surface of COF-TD via in situ reduction of silver ions. Various characterization results confirmed the successful synthesis of flower-like AgNPs@COF-TD with surface-anchored AgNPs, which could expose more active sites. The as-prepared AgNPs@COF-TD exhibited satisfactory oxidase-mimicking activity with a low Km, which could catalyze the oxidation of colorless 3,3 ',5,5 '-tetramethylbenzidine (TMB) to blue oxidized TMB (oxTMB) with significant absorbance at 657 nm. The introduction of DA not only reduced blue oxTMB to colorless TMB but also competed with TMB for the active sites of AgNPs@COF-TD, making a significant decrease in absorbance. Therefore, a label-free sensing strategy based on the oxidase-mimicking activity of AgNPs@COF-TD was successfully established for the colorimetric detection of DA, with a detection limit down to 0.2 mu M. The practical applicability of this assay was further validated by analyzing DA-spiked human urine samples. This work provided a facile approach to prepare COF-based nanozymes, and demonstrated their great potential in colorimetric detection and bioanalysis.

Apoptosis has gained increasing attention in cancer therapy as an intrinsic signaling pathway, which leads to minimal leakage of waste products from a dying cell to neighboring normal cells. Among various stimuli to trigger apoptosis, mild hyperthermia is attractive but confronts limitations of non-specific heating and acquired resistance from elevated expression of heat shock proteins. Here, a dual-stimulation activated turn-on T-1 imaging-based nanoparticulate system (DAS) is developed for mild photothermia (& AP;43 & DEG;C)-mediated precise apoptotic cancer therapy. In the DAS, a superparamagnetic quencher (ferroferric oxide nanoparticles, Fe3O4 NPs) and a paramagnetic enhancer (Gd-DOTA complexes) are connected via the N6-methyladenine (m(6)A)-caged, Zn2+-dependent DNAzyme molecular device. The substrate strand of the DNAzyme contains one segment of Gd-DOTA complex-labeled sequence and another one of HSP70 antisense oligonucleotide. When the DAS is taken up by cancer cells, overexpressed fat mass and obesity-associated protein (FTO) specifically demethylates the m(6)A group, thereby activating DNAzymes to cleave the substrate strand and simultaneously releasing Gd-DOTA complex-labeled oligonucleotides. The restored T-1 signal from the liberated Gd-DOTA complexes lights up the tumor to guide the location and time of deploying 808 nm laser irradiation. Afterward, locally generated mild photothermia works in concert with HSP70 antisense oligonucleotides to promote apoptosis of tumor cells. This highly integrated design provides an alternative strategy for mild hyperthermia-mediated precise apoptotic cancer therapy.

Abstract :
本发明具体涉及了一条抗大田软海绵酸的单链抗体的氨基酸序列及其表达载体。所述氨基酸序列包括:一重链可变区的氨基酸序列,其如SEQ ID No.1所示;一轻链可变区的氨基酸序列,其如SEQ ID No.2所示。编码该重链可变区的核苷酸序列如SEQ ID No.3所示,编码该轻链可变区的核苷酸序列如SEQ ID No.4所示。所述单链抗体的核苷酸序列与带有组氨酸标签麦芽糖结合蛋白的pET28a‑MBP‑T7‑6×His表达载体形成了可溶性重组质粒。
