石花菜醇提物成分分析 CSCD PKU

Abstract :

目的:研究石花菜的成分,为其功能食品的研发提供基础数据。方法:采用薄层色谱、液-质谱联用方法分析石花菜醇提物的化学成分的丰富度。通过硅胶柱、高效液相等色谱方法对其进行分离纯化,并用气-质谱联用、核磁结合文献确证化合物结构;并对部分化合物抗氧化活性进行测定。结果:薄层色谱、液-质谱联用分析表明石花菜醇提物中含有丰富的次级代谢产物,且很可能是甾醇类成分。采用气-质谱联用方法从中鉴定到29个化合物(5个为甾醇类化合物),其中棕榈酸(46.76%)、棕榈酸乙酯(13.82%)、胆甾醇(7.20%)、胆甾-4,6-二烯(4.99%)为含量最高的4个化合物。经色谱分离、核磁鉴定出9个单体化合物,分别为胆甾醇(1)、1,2-二醇植烯(2)、帕格甾醇C(3)、6-羟基-胆甾-4-烯-3-酮(4)、胆甾-5-烯-7-酮(5)、胆甾-5-烯-3β,7α二醇(6)、胆甾-5-烯-3β,7β二醇(7)、胆甾-4-烯-6-酮(8)、3-羟乙基-2-甲基-马来酰亚胺(9)。其中,化合物7清除DPPH自由基和ABTS自由基能力最好,其IC50值分别为180μg/mL和12.2μg/mL。结论:首次系统研究了石花菜醇提物的成分,化合物3、8、9为新天然产物。部分甾醇类化合物具有较好的抗氧化活性。

Abstract :

The purpose of this study was to explore the potential of the development and application of Paecilomyces hepiali, a fungus with edible and medicinal value, as a foodborne antioxidant and anti-aging agent. Its n-butanol extract (PHE) from rice cultures was selected for anti-aging experiment because of significant free radical scavenging activity in vitro. In vivo, PHE could significantly prolong the mean lifespan, 50 % survival days, and the maximum lifespan of Drosophila on a high-fat diet. It is amazing that the mean lifespan increased from 19.1 days to 32.9 days, 50 % survival days increased from 15.7 days to 34.3 days, and the maximum lifespan extended from 44.7 days to 52.7 days, when the high-fat female Drosophila model was fed with 10 mu g/mL PHE. Further research showed that PHE reduced the accumulation of peroxide products and increased the activity of antioxidant enzymes. Then, through antioxidant activity tracking, dimerumic acid (compound 1, the IC50 value of 3.4 mu g/mL on DPPH free radicals scavenging activity), 4,5-dihydroxy-3-methoxypentanoic acid (compound 2, new compound), and thymidine (compound 3) were isolated from PHE. It is worth mentioning that dimerumic acid, the major antioxidant compound of PHE (content up to 3 %), was discovered in P. hepiali for the first time. It was concluded that PHE showed excellent anti-aging activity at a very low concentration on fat diet-induced high fat Drosophila melanogaster, and dimerumic acid may be its main material basis. These results indicated that PHE had the potential to be developed as antioxidant and anti-aging agent in the healthcare industry. (C) 2023 Beijing Academy of Food Sciences. Abstract :

On the basis of single factor experiment, the extraction process of Gelidium amansii ethanol extract (GAEE) was optimized by response surface methodology. Then, the GAEE was successively extracted by petroleum ether, ethyl acetate and n-butanol, and the antioxidant and antibacterial activities of different extraction phases were evaluated. The results showed that the optimum extraction conditions were as follows: extraction time was 64 min, ultrasonic power was 350 W, extraction temperature was 55 !, ethanol concentration was 60% and solid-liquid ratio was 1:35. The extraction rate of GAEE was 1.654 g/g, and DPPH radical scavenging rate of GAEE (2 mg/mL) was 58.29% Subsequent experiments proved that the antioxidant and antibacterial activities of different extraction phases were concentration dependent, and the antioxidant and antibacterial activities of ethyl acetate phase were the strongest. IC50 values of ethyl acetate phase against DPPH, ALTS free radicals, and hydroxyl radical were 672.4, 14.1, 599.7 μg/mL respectively. Their minimum in hibitory concentrations (DIC) of ethyl acetate phase against Staphylococcus aureus and Vibrio parahaemolyticus were 156.3, 156.3 μg/mL, and the minimum bactericidal concentrations (DLC) of ethyl acetate phase against Staphylococcus aureus and Vibrio parahaemolyticus were 625, 312.5 μg/mL. © 2023 Chinese Institute of Food Science and Technology. Abstract :

在单因素实验基础上,通过响应面法优化石花菜醇提物的提取工艺.将石花菜醇提物经石油醚、乙酸乙酯和正丁醇依次萃取,研究不同萃取相的抗氧化和抑菌活性.结果表明,石花菜醇提物的最佳提取工艺为:60%乙醇溶液、料液比1:35、提取时间64 min、超声功率350 W、提取温度55℃.在此条件下,石花菜醇提物得率为0.0165 g/g,DPPH自由基清除率为58.29%(质量浓度2 mg/mL).不同萃取相抗氧化和抗菌活性均具有浓度依赖性,其中乙酸乙酯相活性最强,清除DPPH自由基、ABTS+自由基和羟基自由基的IC50值分别为672.4,14.1,599.7μg/mL.对金黄色葡萄球菌和副溶血性弧菌的最小抑菌浓度(MIC)分别为156.3,156.3μg/mL,最小杀菌浓度(MBC)分别为625,312.5μg/mL.

Abstract :

For food preservation, the packaging film needs to have higher antibacterial activity in initial phase and keep longer activity. In this study, cinnamaldehyde (CA) and its sulfobutyl ether-beta-cyclodextrin inclusion complex (CA/S) were used to fabricate fish gelatin antibacterial composite films. The addition enhanced the elongation at break and light barrier property of the films. Film forming solution incorporated with CA and CA/S presented the most excellent inhibition ratio against Pseudomonas aeruginosa, which was 98.43 +/- 1.11% in initial period and still 82.97 +/- 4.55% at 72 h. Further, the packaging solution of gelatin combined CA and CA/S effectively inhibited the growth of microorganisms during preservation of grass carp slices. Especially, the total volatile salt-based nitrogen (TVB-N) did not exceed 10 mg/100 g at the end of storage, indicating that the active coating could obviously extend the shelf life of fish muscle. This work provided a promising food packaging system with antimicrobial and environmentally friendly.

Abstract :

Obesity has become an increasingly serious public health problem. Pancreatic lipase (PL) is identified as a ideal target for the prevention and treatment of obesity. Orlistat, the only approved PL inhibitor (PLI), is a powerful weight loss drug but has many side effects. Therefore, there is an urgent need to discover powerful PLIs with high safety. Protein hydrolysate has been demonstrated to be a treasure trove of PLIs, but recognizing responsible functional peptides from them is like looking for a needle in a haystack. In this work, we synthesized and optimized a PL ligand fishing model (PLLFM) using magnetic nanoparticles (MNPs), then PLLFM was used to quickly fish out potential PLIs from the Cod meat hydrolysate (CMH). Finally, two new PLIs, GSPPPSG and KLEGDLK were identified with IC50 of 0.60 and 1.08 mg/mL, respectively. The Lineweaver-Burk diagram showed that GSPPPSG is a non-competitively dominant mixed-type PLI, whereas KLEGDLK is a competitive inhibitory-type PLI. Moreover, molecular docking suggested that both peptides can stably bind to the key amino acid residues of the PL active site, mainly through hydrogen bonding, hydrophobic, and electrostatic interactions. In general, we not only established a method to rapidly fish out potential PLIs from protein hydrolysate, but also provided safe and efficient lead compounds for the development of novel diet foods or drugs.

Abstract :

Gelidium amansii (GA) is a kind of red alga homologous to medicine and food and is distributed all over the world. Studies on GA are mainly focused on its polysaccharides, with little research on the ethanol extract. The ethanol extract of Gelidium amansii (GAE) was subjected to a reverse-phase column to obtain 7 components. Among them, 100% methanol solution (GAM), enriched with phytene-1,2-diol, exhibited the strongest DPPH free radical scavenging activity (IC50 = 0.17 mg mL(-1)). Subsequently, high-fat male flies (HMFs) were used as a model to explore the antioxidant and anti-aging effects of GAM in vivo. Studies showed that GAM can effectively prolong the lifespan of HMFs. When GAM concentrations were 0.2 and 1.0 mg mL(-1), the average lifespan of HMFs was increased by 28.7 and 40.7%, respectively, while the longest lifespan of HMFs was increased by 20.55% and 32.88%, respectively. Further research revealed that GAM can significantly downregulate the levels of malondialdehyde (MDA) and protein carbonyl (PCO), and can significantly upregulate the levels of catalase (CAT) and total superoxide dismutase (T-SOD). In addition, by analyzing differential metabolites, we found that GAM relieves aging caused by oxidative stress by regulating amino acid, lipid, sugar, and energy metabolism. The GAM group significantly regulated the levels of adenine, cholic acid, glutamate, l-proline, niacin, and stachyose which tend to recover to the levels of the normal diet male fly (NMF) group. In general, our research provides ideas for the high-value utilization of GA and provides a lead compound for the research and development of anti-aging food or medicine.

Abstract :

To study the chemical constituents from the the deep-sea fungus Alternaria sp. F49. Seven compounds were isolated from the EtOAc extract by using silica gel, Sephadex LH-20, ODS and HPLC methods. Based on the spectroscopic analysis, their structures were identified as (8R)-5-O-methyl-orcinotriol (1), orcinotriol (2), α-acety-lorcinol (3), 3'-hydroxyalternariol 5-O-methyl ether (4), altenusiol (5), altenusin (6), and 5'-methoxy-6-methyl-biphenyl-3,4,3'-triol (7). (8R)-5-O-Methyl-orcinotriol (1) is a new phenolic compound which has never been reported in the literature. Compounds 4-7 showed strong DPPH free radical scavenging activity; whereas compounds 1-7 showed strong ABTS free radical scavenging activity. © 2022, Chinese Pharmaceutical Association. Abstract :

Insight is provided into the mechanism of peptide gelation, a self-assembled process of particular interest, due to its wide existence in natural events and broad applications in food and biomaterials field, but hard to control as minor alterations in the peptide sequence. Here, we design a family of pentapeptides and control their selfassembly behaviors, from micro- and nanoparticles to nanofibers and hydrogels. Furthermore, based on the thermodynamics, the gelation kinetics and the ionization status of terminal groups, the peptide assembly goes through five stages as varying the pH value and the assembly structures are highly dependent on the charging status of the terminal groups. The gelation rules of these peptides are further investigated. Besides, we also load hydrophobic functional molecule (curcumin) into different self-assembly nanostructures with high encapsulation efficiency and remarkable thermal and light protection on curcumin. These peptides assemblies with tunable structures, encapsulation ability and photothermal stabilization could become promising candidates for hydrophobic function factor delivery in food industry.
