Dynamic pH junction-sweeping for on-line focusing of dipeptides in capillary electrophoresis with laser-induced fluorescence detection SCIE

Abstract :

In this paper, we developed a simple and effective on-line focusing technique combining dynamic pH junction and sweeping by capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection. Dynamic pH junction-sweeping is defined when the sample has a different buffer pH (dynamic pH junction condition) and is devoid of micelles (sweeping condition) relative to the background electrolyte (BGE). This hyphenated focusing mode was applied to the sensitive and selective focusing of four dipeptides: Tyr-Phe, Tyr-Leu, Trp-Gly, and Ala-Gln. Picomolar detectability of these dipeptides by CE-LIF detection was demonstrated through effective focusing of large sample volumes (up to 39% capillary length) using the dual pH junction-sweeping focusing mode. 25 mmol L-1 sodium dihydrogen phosphate, pH 2.5 was used as the sample matrix, and 100 mmol L-1 borate, 21 mmol L-1 sodium dodecylsulfate (SDS), 16 mmol L-1 Brij35, pH 9.0 as the background solution (BGS). The concentration detection limits (S/N = 3) of the four dipeptides were in the range of 1.0-5.0 pmol L-1. The developed method has been successfully used for the determination of dipeptides in human serum samples.

Abstract :

A method was developed for the chiral separation of dipeptides with two chiral centers. Although all analyzed peptides contain chromophores and they can be detected by UV-adsorption detection without any derivatization, the chiral separations were not achieved for non-derivatized peptides using the same chiral selectors and experimental conditions. In this paper, these dipeptides were precolumn derivatized by using naphthalene-2, 3-dicarboxyaldehyde as a tagging reagent. In the presence of 2-hydroxypropyl-beta-CD and sodium deoxycholate, naphthalene-2,3-dicarboxyaldehyde-tagged dipeptide enantiomers were well resolved by MEKC. Good separation of all enantiomers was obtained within 14 min. The proposed method was applied to chiral separation of dipeptide enantiomers in spiked human serum samples.

Abstract :

Microcrystalline Ni (MC-Ni), ultrafine grained Ni (UFG-Ni) and nanocrystalline Ni (NC-Ni) with mean grain sizes of 2 mu m, 120.7 nm and 22.4 nm, respectively were prepared by a pulsed electrodeposition method. The room-temperature one-way tensile tests show that the yield strength and the ultimate tensile strength of the three kinds of Ni deposits increase with decreasing of the grain size. The ductility of UFG-Ni decreases to 7.9% compared with 11.1% of the MC-Ni, while the UFG Ni shows a higher uniform elongation than MC-Ni. However, the ductility of NC-Ni is only 4.1%.

Abstract :

A CE with LIF detection was developed for the separation and determination of three bradykinin (BK) metabolites: BK2-9, BK1-7 and BK1-5. BK fragments were derivatized with 5-(4, 6-dichloro-s-triazin-2-ylamino) fluorescein before CE-LIF analysis. Eighty millimolar of Tris-H3PO4 (pH 9.0) was selected as the derivatization media. Three BK fragments were baseline separated within 10 min by using 0.2 M TAPS-Tris buffer (pH 8.5) as the running buffer. Meanwhile we have also developed a simple, quick, and sensitive on-column transient ITP preconcentration for CE-LIF detection of three BK fragments. Ten millimolar of Tris-HCl (pH 9.0) was chosen as the leading electrolyte and 0.2 M TAPS-Tris (pH 8.5) containing 10 mM 1-butyl-3-methylimidazolium tetrafluoroborate as the terminating electrolyte and also served as the running buffer during CZE separation. By using this transient ITP coupled with CE-LIF, concentration detection limits (S/N= 3) for BK2-9, BK1-7 and BK1-5 were 0.3, 0.1 and 0.1 pmol/L, respectively. This method has been applied to the assay of human saliva and plasma samples with satisfactory results.

Abstract :

采用氨基磺酸镍体系镀镍液代替瓦特型镀镍液,应用脉冲电沉积技术,制备了平均晶粒尺寸为16.7 mn的纳米镍.在静拉伸应变速率范围(5×10-5~10-2s-1)内,纳米镍的强度和塑性均随应变速率的增加而增加,断裂形式表现为韧窝韧性断裂,获得的最高断裂强度和最大断裂延伸率分别为1332MPa和5.31%.在退火温度为100、150和200℃保温1 h后,室温应变速率为10-3s-1时,纳米镍的强度和塑性随退火温度的升高而显著下降,原因为退火过程中晶内硫元素向晶界强烈偏聚引起材料变脆,断裂形式表现为晶间脆性断裂.XRD和TEM的观测结果表明,纳米镍在250℃退火时,晶粒发生显著长大,硬度快速下降.热分析仪测得的放热峰表明,275℃以后,大量的纳米晶粒发生异常长大.

Abstract :

A new method for the determination of the peptide hormones and their fragments by capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection and transient pseudo-isotachophoresis (pseudo-tITP) preconcentration was established in this study. The LIF detector used an argon ion laser with excitation wavelength at 488 nm and emission wavelength at 535 nm. Fluorescein isothiocyanate (FITC) was used as precolumn derivatization reagent to label cholecystokinin tetrapeptide (CCK-4), neurotensin (NT), neurotensin hexapeptide (NT8-13), and neurokinin B (NKB). Borate (10 mmol/L, pH 9.0) was selected as derivatization medium to get the high efficiency. When the addition of 70% (v/v) methanol and 1% (m/v) sodium chloride (NaCl) to the sample matrix, and with borate buffer (110 mM, pH 9.5) and 20% (v/v) methanol as running buffer, a preconcentration based on the pseudo-tITP afforded 100-fold improvement in peak heights compared with the traditional hydrodynamic injection (2.3% capillary volume). The detection limits (signal/noise =3) based on peak height were found to be 0.04, 0.1, 0.2, and 0.08 nmol/L for NT8-13, NT, NKB, and CCK-4, respectively. The method was validated and applied to qualitative analysis of NT and NT8-13 in human cerebrospinal fluid sample. (c) 2008 Elsevier Inc. All rights reserved.

Abstract :

A CE with LIF detection was developed for separation and determination of bradykinin (BK)-related peptides, such as BK, kallidin (Kal), and neurokinin A (NKA). BK-related peptides were derivatized with FITC prior to CE-LIF analysis. Sodium borate 10 mmol/L at pH 9.5 was selected as derivatization media in order to get the high efficiency. Three peptides were baseline-separated within 10 min by using 110 mmol/L sodium borate-sodium hydroxide solution at pH 10.0 as the running buffer. Concentration detection limits (S/N = 3) for BK, Kal, and NKA were 0.08, 0.5, and 0.2 nmol/L, respectively. Meanwhile we have also developed a simple, quick, and sensitive large-volume sample stacking (LVSS) technique for CE-LIF detection of BK, Kal, and NKA. By using this stacking technique, the detection limits (S/N = 3) for BK, Kal, and NKA were 0.02, 0.05, and 0.04 nmol/L, respectively. This method has been applied to the assay of human saliva and cerebrospinal fluid with satisfactory results.

Abstract :

采用毛细管电泳安培检测法对厚朴酚及和厚朴酚的分离测定进行了详细研究.工作电极为0.3 mm碳圆盘电极,30 mmol/L、pH 10.0的硼砂-氢氧化钠为运行缓冲液,分离电压21 kV,检测电位0.95 V.在最优化实验条件下,两待测物在6 min以内完全基线分离.厚朴酚及和厚朴酚的线性范围分别为0.2~50 μg/mL和0.4~60 μg/mL,检测限分别为0.06和0.2 μg/mL.应用于中药厚朴以及中成药霍香正气水和保济丸中厚朴酚及和厚朴酚的检测.

Abstract :

沙丁胺醇(Sal)是选择性β2受体激动剂的一种,可作为畜禽的促生长剂,但Sal易在动物脏器中积聚残留,通过食物链进入人体,严重危害人类健康.迄今已报道的沙丁胺醇的检测方法有高效液相色谱法[1]、气相色谱法[2]、酶联免疫吸附法(ELISA)[3,4]等.然而这些方法都具有检测过程烦琐,检测时间长等缺点.本研究建立一个快捷灵敏地检测Sal的Nation修饰玻碳电极方波溶出伏安法的测定体系,并且探讨了Sal在Nation修饰电极上的电极反应机理.
