Abstract:

以课题组从海鳗肠道内容物中筛选获得的洋葱伯克霍尔德菌ZF-16为试验菌株,采用单因素试验和均匀设计试验对其培养基成分和发酵条件进行研究,得到优化的产酶条件为:玉米淀粉0.5%,蛋白胨3.0%,橄榄油10.0%(体积分数),MgSO_4·7H_2O 0.05%,K_2HPO_40.1%,培养基初始pH 6.0~7.0;培养温度30℃,摇床转速120r/min,装液量50 m L/250 m L,接种量2%(体积分数),接种龄18 h,发酵时间68 h。在此条件下发酵,洋葱伯克霍尔德菌ZF-16的脂肪酶活力为72.59 U/m L,较优化前(12.71 U/m L)提高了4.71倍。
