Abstract:

An O-cyanoethylchitosan modified open tubular capillary electrochromatography column was prepared. The optimized monomer concentration was 7.5 mg/mL. The stability and reproducibility of the column were characterized by electroosmotic flow. The relative standard deviation (RSD) is less than 0.2% (run-to-run, n=3), 0.6% (day-to-day, n=3), 1.0% (column-to-column, n=3), 3.4% (batch-to-batch, n=3). In addition, the structure and morphology were characterized by infrared spectroscopy (IR) and scanning electron microscopy (SEM). Four kinds of nucleotides were separated successfully on the modified capillary electrochromatography column, and the column efficiency is between 37, 000 to 140, 000 plate/m. The results show that the preparation method of the open-tubular capillary is simple, effective and stable.
