以课题组从龙须菜中分离得到的产琼胶酶弧菌Ag-1为试验菌株,对其产酶培养基和发酵条件进行研究,得到优化后的培养基配方和培养条件为:琼脂0.2%,蛋白胨0.3%,酵母膏0、8%,NaCl 4.0%,MgSO4 0,6%,CaCl2 0.2%;培养基初始pH 6.5,接种量1%(体积分数),摇床转速240 r/min,装液量50 mL/250 mL,发酵温度33℃,发酵周期24 h.在此条件下做发酵产酶试验,琼胶酶活力达45.51 U/mL,较优化前的7.0 U/mL提高了5.5倍.
